This experiment was carried out to exploit effective and easy made sub-culture medium for production of vigorous seedlings of Cymbidium kanran derived from in vitro cultured rhizome, and to observe the effects of salt strength of Murashige and Skoog (MS) medium, banana homogenate and apple juice on plantlets growth of the orchid in vitro.
Number of new shoot and fasciculate leaf was increased in MS, 1/2MS and 1/3MS medium. Higher de-differentiation ratio was marked in MS or 5g/ℓ Kampsal medium; however, none was observed in 3g/ℓ Kampsal medium containing 15g/ℓ apple juice.
Although root formation was inhibited in Kampsal medium, 1g/ℓ Kampsal and 3g/ℓ Hyponex medium were effective for production of vigorous plantlets of Cymbidium kanran in vitro. The most effective sub-culture medium for the Cymbidium plantlet was 3g/ℓ Kampsal medium containing 20g/ℓ apple juice, 3g/ℓ peptone, lg/ℓ activated charcoal, 30g/ℓ sucrose and 7g/ℓ agar.
